The cell culture medium is sometimes supplemented with antibiotics in appropriate concentrations, which can effectively prevent microbial contamination. At present, the most commonly used antibiotic is penicillin-streptomycin mixed solution (100× double antibody). Penicillin-streptomycin-amphotericin B Solution (100× triple resistance) is specially used for cell culture, and can be directly added to cell culture Solution after filtration. The penicillin-streptomycin-amphotericin B mixed solution (100× triple antibody) contained 10kU/ml penicillin, 10mg/ml streptomycin and 25µg/ml amphotericin B. The solution was prepared with 0.9%NaCl or PBS, and the recommended working concentrations of penicillin, streptomycin and amphotericin B in cell culture medium were 100U/ mL, 0.1mg/ mL and 0.25µg/ mL, respectively. A package of 100ml penicillin-streptomycin-amphotericin B mixture (100× triple antibody) can be used to prepare 10L cell culture medium.

 

Penicillin-streptomycin-amphotericin B mixture (100 x triple antibody) can be used in one of the following two ways:

 

1. Add directly to sterile cell culture medium: add penicillin-streptomycin amphotericin B mixed solution (100× tricyclic antibody) at the ratio of 5ml per 500ml cell culture medium, and mix well for use.

 

2. Add when preparing cell culture medium, and then filter for sterilization: When preparing cell culture medium, add penicillin-streptomycin - amphotericin B mixed solution (100 x triple antibody) at the ratio of 10mL to each 1L cell culture medium, and filter for sterilization after preparation.

 

Matters needing attention:

 

1. Minimize the number of repeated freezing and thawing to avoid efficiency decline.
